Cam Chain Timing

So I was rotating the engine on my 2006 ninja 250 without the camshaft bearing caps (had them removed to check the gasket) and I'm worried that I might have caused it to skip a tooth and messed up the cam timing. What's the correct way to make sure it's timed correctly and safe to ride? I read on some other forum that the way to check it is by ensuring that the intake and exhaust marks on the cam gears line up with the valve cover gasket and the engine is at "2T". I also counted the chain link pins and they add up to 33. Is this correct ? Thanks for any help.
