That looks like a drain hole to let moisture out of the muffler.
With it idling you can cover the exhaust outlet with the sole of your shoe (or have a helper do it) and listen for exhaust escaping. If it's been leaking for a while there may also be deposits around the area.
You won't get any reaction or RPM change by spraying anything on the exhaust, that only works for finding intake leaks. Not sure I'd spray anything on a hot exhaust system.
