Anything other than Prime is fine, as long as your vacuum-actuated petcock is working properly. It closes when there's no engine vacuum applied. Turn it Off just to be safe.
On models with Prime, that will allow fuel to flow without any vacuum applied, for refilling the carbs after they've been drained. On models without Prime, you'll need to apply vacuum to the petcock (either manually or by cranking the engine for a little bit) to allow the carbs to refill.
