Petcocks don't create vacuum. They're just a valve to control fuel flow from the tank to the carbs. You can rig up a temporary tank of some sort and run a line from it to the carbs to see if your problem is the petcock or the carbs.
Or if this is an aftermarket vacuum petcock you can just suck on its vacuum line with your mouth to test its flow. And if it's not a vacuum petcock, just open it and see if it flows fuel.
My guess is that something is wrong in the carbs, but that's just speculation.
