One cylinder not firing
Hey guys I have a 2005 ninja 250 the California model with 6600 miles on it. I'm struggling getting the bike started. It has new plugs that have spark, timing has been checked and is good, cleaned carbs, and replaced battery, seems to have good compression from both cylinders. When I unplug the boot from the left side plug it starts and idles rough but I have no throttle response at all. When both plug boots are on it struggles to start up and seems to backfire. Has anyone had this issue or have any advice? Thanks

Just for fun, try switching the plug wires.
Welome to the board.

Welcome, Adam !!!
Do you refer to the timing of the valves?

Did you verified the gap of each valve was within specs?

It is easy if you have a shaded place to work and some basic tools. This is how to: http://faq.ninja250.org/wiki/How_do_..._the_valves%3F http://n4mwd.blogspot.com/2013/06/ov...ts-during.html Backfiring may be caused, among other things, by some loose electrical connector in the ignition circuit: http://faq.ninja250.org/wiki/Wiring_...s_%26_diagrams Having no throttle response at all sounds like broken diaphragms of the carbs or incorrectly installed needles: http://faq.ninja250.org/wiki/How_do_...n_the_carbs%3F

A bonus: https://www.ninjette.org/forums/showthread.php?t=151139 Those are basic things to check, just to achieve at least combustion inside the chambers: air, fuel, compression and a healthy reliable spark.

So with the now throttle response I pulled the diaphragms out and the spring on that side was bent and kind of screwed up could that be the issue there?

Those diaphragms are activated by the vacuum of the engine and move those tapered needles up and down, which allows more or less fuel into the stream of air being drawn into the cylinders. This is how the CV (constant velocity) carburetors work:
