Bike won't start, cleaned carbs, new gas, backfires whats next?

I was having a rough/staling idle so i cleaned the carbs, and can't get the bike to start back up.

-the carbs work flawlessly in a different bike
-Gas is brand new 87 octane-today, gas was added by using the prime option for a little bit then set to "on", I tried draining the bowls just in case i flooded them and still no luck
-The bike is backfiring every so often so i have spark(right?), but I will replace spark plugs anyway on Friday.
-The bike inches forward a bit when in gear and hitting the starter (so i think the starter motor is still good)
-The battery is fully charged before trying tests (battery tender has green light)
-I can say I checked and re checked all hoses and wires but youll say check it again :\

So what would be my next steps to get this bike to fire?
Is it an issue with timing of the spark and the fuel or lack of?
Is there a way to test the vacuum system?

Do you have the air filter on it and buttoned up?

Air filter in, boots are lined up on all 4 sides and tightened down

Are you using choke? If so, I would turn it off after you hear any signs of life at all or after cranking for 5-10 seconds max. If it's on too long it may flood. Did you remove the idle mixture screw caps and change the settings? You may need to try different adjustments around 2 turns out.

Also try opening the throttle slightly while cranking with minimal or no choke.

Looking at the color of the plugs may give you some idea of a rich or lean condition.

You will most likely need to have the battery on a small charger if you are doing any amount of cranking.

yes tried choke on and off

will try your suggestions and report back tomorrow

I did remove the caps of the idle screws to clean the, and returned them to stock settings

Hey guys i replaced the plugs and it fired right up , no idea how they went bad just by pulling carbs off?

Csmith I tried adjust idle screws to no avail, waited for the plugs before doing anything else, then it turns out I didn't need to do anything else!
